ABSTRACT
Background and purpose:Aberrant expressions of microRNAs (miRNA, miR)are reported in various cancers and may associate with cancer occurrence, development, invasion and metastasis, thereby functioning as either tumor suppressors or oncogenes. This study attempted to observe the expression of miR-214 in pancreatic cancer and to explore its clinical signiifcance.Methods:Real-time PCR was used to detect the miR-214 expressions between pancreatic cancer tissues and matched adjacent tissues. The correlations of miR-214 expression with clinic-pathological features and clinical prognosis were analyzed.Results:MiR-214 expression was up-regulated in 69.4%(25/36) of tumor tissue specimens. The relative expression level of miR-214 was signiifcantly higher in tumor tissues than in matched adjacent tissues (3.45vs 1.52,P<0.01). Higher miR-214 level was strongly associated with T3-T4 stage (P=0.018). The Kaplan-Meier analysis revealed that patients with higher expression of miR-214 had a shorter survival time (P=0.032).Conclusion:The expression of miR-214 is associated with clinic-pathological features and patient’s clinical prognosis, so it may be used as a potential diagnostic biomarker and a prognostic predictor in patients with pancreatic cancer.
ABSTRACT
Objective To investigate the effect of Gum mastic combined with gemcitabine on human pancreatic carcinoma BxPc-3 cells and its mechanism.Methods Cell proliferation and apoptosis were examined using the methyl thiazolyl tetrazolium assay and propidium iodine staining,respectively.After BxPc-3 cells were treated with different concentrations of Gum mastic and gemcitabine,the expression of NF-κB p65 subunit,IkB,Bcl-2 and Bax proteins was detected by Western blot.BxPc-3 cells were injected subcytaneously into nude mice to establish pancreatic xenograft tumors,and the changes of tumor volume were monitored.Results Compared to either single agent,treatment with Gum mastic (40 mg/L) combined with gemcitabine (10 mg/L) for 72 h signi cantly inhibited the proliferation of BxPc 3 cells (P＜0.01).Its rate of apoptosis(45.13±4.01)was more than Gum mastic,gemcitabine(P＜0.01) and control group (5.07 ± 1.37,P＜ 0.01).When cells were treated with gemcitabine in combination with gum mastic in human pancreatic carcinoma BxPc-3 cells for 48 h,the IκB level was increased,whereas NF-κB activation was blocked; the expression of Bax protein was substantially increased,but Bcl-2 protein was down-regulated; gum mastic or combined with gemcitabine could significantly inhibit the growth of pancreatic xenograft tumors (P ＜ 0.05).Conclusions Gum mastic could effectively strengthen the sensitivity of human pancreatic carcinoma BxPc-3 cells to gemcitabine.It may inhibit the expression of NF-κB p65 subunit and Bcl-2 proteins and increase the expression of IκB and Bax proteins.
ABSTRACT
Objective To evaluate the value of the methods of diagnosis and surgical intervention of liver cavernous hemangioma(LCH).Method The clinical data of 31 cases undergoing surgical treatment for LCH were retrosprectively analysed.Results Out of 31 cases,21 has only one focus each,the others has 2 or more foci each.The LCH were located only in the right lobe in 19 cases,only in the left in 9,and in both in 3.The diameter of the tumors were 10 cm(n=6).The correct diagnostic rate of BUS, CT, and Nuclear Imaging(NI) was 90.3%,95%,and 100%,respectively.Hepatectomy were performed on all 31 patients.There was no motality,and the complicative rate was 19.4%(6/31).Conclusions BUS and enhanced CT are important in discovering,diagnosing and differentiating LCH.We think that hepatectomy is the saftest and effective method for patients with LCH over 5 cm in diameter,especially for those with symtoms.
